<strong>مقدّمه</strong>: بیماری های قلبی-عروقی و سکته، دو عامل مهم و مسئول حدود 25 درصد مرگ و میر در سراسر جهان تا سال 2030 هستند.وابستگی هایپرکلسترولمی به این دو عامل، آن را به یکی از مهم ترین عوامل شرکت کننده در مرگ زودرس مرتبط با بیماری های قلبی و عروقی تبدیل کرده است.<br>
هایپرکلسترولمی خانوادگی(<span dir="LTR">FH</span>) یک بیماری اتوزومی غالب است که از طریق میزان <span dir="LTR">LDL</span> در پلاسمای خون شناسایی می شود. تشخیص بالینی این بیماری بر پایه ی تاریخچه ی شخصی و خانوادگی، یافته های معاینات فیزیکی و اندازه گیری میزان غلظت کلسترول بالا انجام می شود.بیشترین علل تک ژنی ایجاد کننده ی<span dir="LTR">FH</span>، ایجاد جهش در یکی از سه ژن <span dir="LTR">LDLR</span>، <span dir="LTR">APOB</span> و <span dir="LTR">PSCK9</span> هستند که حدود 90درصد آن ها در ژن <span dir="LTR">LDLR</span> بوجود می آید و تاکنون بیش از 1700 جهش مختلف در آن شناسایی شده است.<br>
<strong>روش کار</strong>: در این مطالعه ی مروری، جستجو در پایگاه های اطلاعاتی الکترونیکی و علمی <strong><span dir="LTR">NCBI</span></strong>، <strong><span dir="LTR">Science Direct</span></strong>، <strong><span dir="LTR">Google Scholar</span></strong>، انجام شد و مقالات معتبر مرتبط با موضوع مورد استفاده قرار گرفت.<br>
<strong>یافته ها</strong>: این مقاله بهمرور یافته های نوین در زمینه ی شایع ترین بیماری ژنتیکی جهان، یعنی هایپرکلسترولمی خانوادگی، عوامل ژنتیکی مسبّب این بیماری، درمان، و تشخیص بالینی و ملکولی این بیماری می پردازد.<br>
<strong>نتیجه گیری:</strong><br>
با وجود شیوع بالای بیماری هایپرکلسترولمی خانوادگی در جهان،خوشبختانه این بیماری قابل تشخیص بوده و درمان آن امکان پذیر است. تشخیص زودهنگام این بیماری می تواند خطر ابتلا به بیماری های قلبی-عروقی و مرگ زودرس ناشی از آن را کاهش دهد. از آنجایی که معیارهای بالینی ممکن است تمام بیماران مبتلا را شناسایی نکند، بهتر است از تشخیص ملکولی و آزمایش ژنتیک در استراتژی های غربالگری استفاده شود.<br>
<strong>Introduction: </strong>Cardiovascular disease and stroke are the two major causes of about 25% mortality worldwide by the year 2030. The dependency of Hypercholesterolemia on these two factors has made it one of the most important contributors to early death associated with cardiovascular disease. Family Hypercholesterolemia (FH) is a predominant autosomal disease detected by the level of LDL in the blood plasma. Clinical diagnosis of this disease is based on clinical and family history, clinical findings, and high cholesterol concentration. More commonly, the single-gene causes of FH are the mutations in one of the three LDLR, APOB, and PSCK9 genes. About 90% of them are in the LDLR gene, and more than 1700 mutations have been identified in this gene.<br>
<strong>Methods: </strong>In this study, the scientific and electronic databases including, NCBI, Science Direct, and Google Scholar were used. Authentic articles related to the topic also were used.<br>
<strong>Results: </strong>This article reviews new findings on the most common genetic disease in the world, namely familial hypercholesterolemia, genetic causes of the disease, treatment, and clinical and molecular diagnosis of the disease.<br>
<strong>Conclusion: </strong>Despite the high prevalence of familial hypercholesterolemia in the world, fortunately, the disease is detectable and can be treated. Early diagnosis of the disease can reduce the risk of cardiovascular disease and early death. Since clinical criteria may not identify all patients, it is better to use molecular diagnosis and genetic testing in screening strategies
